Drug intelligence / Profile preview
[68Ga]Ga-THP-Pam is a gallium-68 labeled bisphosphonate positron emission tomography (PET) radiotracer developed for imaging vascular calcification and bone-related conditions. It consists of the clinically used aminobisphosphonate pamidronate (Pam) conjugated to the rapid-chelating tris(hydroxypyridinone) (THP) ligand, which allows for simple, one-step, room-temperature radiolabeling with gallium-68. Unlike [18F]NaF, which is highly selective for hydroxyapatite (the main component of bone mineral), [68Ga]Ga-THP-Pam exhibits a broad affinity for various calcium minerals (such as whitlockite and other non-hydroxyapatite calcium salts) implicated in extraosseous and vascular calcification. This broad mineral affinity makes it a highly sensitive diagnostic agent for the early detection of vascular calcification and microcalcifications.
